Akt deficient mice reduced high tidal volume-induced Akt activation. Representative photomicrographs (x400) with phospho-Akt staining of lung sections via immunohistochemistry were from 5 days of bleomycin-treated wild type or Akt+/- mice ventilated at tidal volume of 6 ml/kg or 30 ml/kg for 5 hours with or without pretreated with 2 mg/kg of PD98059 subcutaneously for 30 minutes (n=6 per group). (a) Control wild-type mice; (b) Control Akt+/- mice; (c) VT 6 ml/kg wild-type mice; (d) VT 30 ml/kg wild-type mice; (e) VT 30 ml/kg wild-type mice pretreated with PD98059; (f) VT 30 ml/kg Akt+/- mice. A dark brown DAB signal indicates positive staining for S100A4 in the lung epithelium or interstitial while shades of bluish tan signifies non-reactive cells.
